Many basic fluorescence plate readers are not equipped to run Alpha assays. To ensure the best possible results, we recommend using our VICTOR® Nivo™, EnSight®, or EnVision® multimode plate reader, each of which comes with pre-set Alpha settings. You can use the same generic instrument protocol for AlphaScreen and AlphaLISA assays.
If you are using a plate reader from another company, we recommend that you check to see if the manufacturer has validated the instrument for an AlphaScreen or AlphaLISA assay. This information is usually found on the company’s website. PerkinElmer has not validated other instruments, and we would recommend that you speak to the instrument manufacturer for information on the recommended settings to use.
Already equipped with a PerkinElmer reader? You are all set! Our reader comes with pre-set Alpha settings ensuring you’ll get the most out of your assay with your reader.
Alpha technologies require excitation at 680 nm and reading emission at 520-620 nm (for AlphaScreen) or at 615 nm (for AlphaLISA). A laser excitation source is usually needed, as an intense light source is required to generate the signal for fast and sensitive readout. Xenon Flashlamp based systems and luminometers will not work for Alpha assays.

LANCE Ultra assays require a microplate reader that is able to perform time-resolved fluorescence. We recommend filter-based fluorometers, though monochromator instruments can be used taking into account possibly decreased assay sensitivity. We recommend using our VICTOR® Nivo™, EnSight®, or EnVision® multimode plate reader with the appropriate filters for these assays.
If you are using an instrument manufactured by another company, we recommend that you contact their technical support team to see if they have optimized settings, filter recommendations, etc. for a LANCE TR-FRET assay.
Use an excitation wavelength of 320 or 340 nm to excite the LANCE Europium chelate. We recommend you read this assay in dual emission mode, detecting both the emission from the Europium donor fluorophore at 615 nm, and the acceptor fluorophore (at 665 nm for APC, ULight dye, and Alexa Fluor 647). The raw FRET signal at 665 nm can be used to process your data. The ability to analyze the signal at 615 nm can be helpful when troubleshooting.

DELFIA assays require a plate reader that is capable of TRF (time-resolved fluorescence) detection. This is a time-resolved fluorescence intensity assay. We recommend using our VICTOR® Nivo™, EnSight®, or EnVision® multimode plate reader.
We recommend a time-resolved filter-based fluorometer for DELFIA Europium assays. Very sensitive time-resolved spectrofluorometers may be suitable for certain assays. Most plate readers are capable of TRF-type assays – a flash lamp is needed for TRF analysis, which is an option on most plate readers. You could also use laser excitation on an appropriate EnVision instrument.
When using DELFIA Europium, we recommend generic default instrument settings for all DELFIA assays.
We always recommend that you check with the manufacturer of your particular instrument to see if they have additional recommendations for DELFIA assays.
